Experimental Studies on The Source of Endogenous Pyrogenic Substance

Abstract
The study was carried out to ascertain that normal tissue could be a source of endogenous pyrogenic substance and, further, that the mechanism of heat production by saline extract of normal tissues.
1. Normal rabbits were injected with saline tissue extracts of the heart, liver, spleen, adrenal body, and
kidney, excised from the rabbits after sacrifice by exssnguination and tested their fever responses. The results showed all were positive in pyrogenic activity
2. With successive daily injections of saline tissue extracts into the normal rabbits, the author observed whether tolerance could be obtained or not, by the inoculated animals. It was resulted that tolerance could not be obtained by them.
3, In order to ascertain whether saline tissue extracts act directly on the thermoregulatory centers or on the heat-producing organs and tissues, or act indirectly on them with secondary endogenous pyrogenic substance released into the blood stream, normal rabbits were injected the sera from the blood of donor rabbits inoculated by saline tissue extracts and tested their fever responses.
It was concluded that no secondary endogenonus pyrogenic substnce was released into the blood stream of the inoculated animals.
4. Saline extracts of normal tissues were injected into:
A. The animal (function of the C. N. S- inhibited by barbital sodium and sepamin) B. The animal (lower cervical section)
C. The animal (bilaterally spianchnicectomized at the level just below the diaphragm) D. The animal(bilaterally vagotomized at the neck)
E. The animal with thyroideetomy.
The result showed that the normalities of the thermoregulatory centers and the thyroid gland were not necessary for mechanism of heat production by the extracts. The author confirmed the mechanism of heat production by the extracts was peripheral.
5. In order to characterize the nature of pyrogenic substance, which was supposed to exist in the saline
extracts of normal tissues, biochemical analysis was performed by several methods.
The results showed that pyrogenic substance mainly belonged to globulin fraction of the extrats,
6. Through these experiments, the author confirmed that normal tissue could be a source of endogenous pyrogenic substance.
